چکیده

Darters (Percidae: Etheostomatini) offer attractive opportunities for analysis of the evolution of reproductive barriers and the molecular basis of fertilization. Here we report some basic characteristics of the sperm of darters, and, to our knowledge, the first report of sperm associated -L-fucosidase in fishes. Fluorometric enzyme assays using 4methylumbelliferyl-alpha-L-fucopyranoside [4-MU-fuc] as a substrate provided direct quantification of enzyme activity in both the soluble and cellular fractions of milt from all six species tested. Further analysis using SDS-PAGE and Western blotting revealed that fish sperm and milt contain -L-fucosidase isoforms with molecular weights similar to those reported for human semen. Fertilization of darter ova by conspecific sperm, as assessed by development of the embryo to the 4 cell stage, was inhibited by pretreatment of sperm with the fucosidase-specific competitive inhibitor DFJ, implicating the sperm fucosidase in fertilization and/or early embryogenesis. Identification of sperm associated -Lfucosidase in darters is consistent with the importance of carbohydrates during fertilization.

Effect of successive milt collection on sperm quality and reproduction of wild and cultured endangered caspian brown trout, Salmo trutta

In the present study, the effect of successive milt collection was investigated on spermatological parameters and reproduction successes of wild and cultured endangered caspian brown trout. The milt collection was done 3 times with two weeks intervals. After milt collection, amount of milt was allocated for milt quality evaluation and reminders used for fertilization. According to results, in w...

The effect of phylogeny on interspecific body shape variation in darters (Pisces: Percidae).

We conducted a geometric morphometric analysis of interspecific body shape variation among representatives of 31 species of darters (Pisces: Percidae) to determine whether there is evidence of a phylogenetic effect in body shape variation. Cartesian transformation grids representing relative shape differences of individual species and subspecies revealed qualitative similarities within most tra...

Life History of the Yazoo Darter (percidae: Etheostoma Raneyi), a Species Endemic to North-central Mississippi

The Yazoo darter, Etheostoma mneyi, is restricted to tributaries of the Little Tallahatchie and Yocona Rivers (Yazoo River drainage) in north-central Mississippi. The species inhabits small, clear streams, many of which are spring-fed, and have a variety of substrate types including silt, clay, sand and gravel. In Morris Creek, a second-order stream in Lafayette County, Mississippi, E. runeyi w...
